Before Repeating the Experiment

Before repeating the entire experiment, ask yourself:

โœ“ Enough protein loaded?
โœ“ Transfer confirmed?
โœ“ Correct secondary antibody?
โœ“ Fresh ECL?
โœ“ Housekeeping protein detected?

If all five answers are yes, you’ve ruled out the most common technical issues. At that point, repeating the experiment is usually more productive than continuing to optimize the same blot.
